Part:BBa_K5036002
NLS
Part Description
It is a nuclear localization signal which is a short sequence of amino acids found in certain proteins that acts as a zip code, directing those proteins to the nucleus of a cell. In our model it is attached to dCas9(C) in the first chain of dCas9(C)-TF-synVEGFR1 receptor.
Usage
This part is attached to dCas9(C) so after activation of the receptor and dimerization of the two domains of dCas9, NLS will direct the (dCas9-VP64-GAL4- UAS Trans CMV enhancer) complex into the nucleus.
this figure illustrates the structure of nuclear localization signal (NLS) which is attached to dCas9(c) in dCas9(C)-TF-synVEGFR1 receptor's first chain. .
Experimental Characterization
we digested dCas9(C)_NLS-Syn-VEGFR-1 grafetd with NLS and performed gel electrophoresis to detect the digested fragment.
This figure illustrates the digested dCas9(C)_NLS-Syn-VEGFR-1 prior to insert ligation .
